İşlevi
Bu gen tarafından kodlanan protein, dizilim bakımından ve muhtemelen işlev açısından retinoblastoma 1 (RB1) geninin ürünüyle benzerdir. RB1 gen ürünü, hücre döngüsünün düzenlenmesinde rol oynadığı düşünülen bir tümör baskılayıcı proteindir; S'den M fazına geçiş sırasında fosforile edilirken, hücre döngüsünün G1 fazında defosforile edilir. Hem RB1 proteini hem de bu genin ürünü, adenovirüs E1A proteini ve SV40 Large T-antijeni ile kompleks oluşturabilir; SV40 Large T-antijeni ise her iki proteinin yalnızca fosforile edilmemiş formuna bağlanır. Ayrıca, her iki protein de promotörlerinde E2F bağlanma bölgeleri bulunan hücre döngüsü genlerinin transkripsiyonunu inhibe edebilir. RB1 proteini ile olan dizilimsel ve biyokimyasal benzerlikleri nedeniyle, bu gen tarafından kodlanan proteinin de bir tümör baskılayıcı olabileceği düşünülmektedir.
